Sofosbuvir
Tacrolimus
Summary:
Coadministration of tacrolimus (5 mg single dose) and sofosbuvir (400 mg single dose) was studied in 16 subjects. Sofosbuvir Cmax decreased by 3% and AUC increased by 13%. GS-331007 Cmax decreased by 3% and there was no change in AUC. Tacrolimus Cmax decreased by 27%, but AUC increased by 9%. No a priori dose adjustment of sofosbuvir or tacrolimus is required. However, the pharmacokinetics of tacrolimus may be impacted by changes in liver function during DAA therapy and monitoring of tacrolimus levels is recommended.
